Emergency services were called to the home just before 7am, after receiving calls to Triple Zero.
Fire crews arrived within minutes to find the two-story home completely engulfed in flames.
Five people were evacuated from the house, while several people suffered smoke inhalation and burns.
Paramedics treated a teenage boy at the scene before taking him to the Royal Children’s Hospital.
A man in his 60s and three children were also taken by ambulance to Werribee Mercy Hospital.
The Victoria Fire and Rescue Service said the fire was under control at around 7.35am.
The house was badly damaged by the fire.
Point Cook is located 22 km southwest of Melbourne’s CBD.
